Protalix BioTherapeutics, Inc.
Protalix BioTherapeutics, Inc. is a publicly traded biopharmaceutical company (NYSE American: PLX; TASE: PLX) headquartered in Carmiel, Israel. The company specializes in the development, clinical testing, and manufacturing of recombinant therapeutic proteins designed to treat rare genetic conditions and metabolic disorders. Protalix is recognized globally as the first entity to achieve regulatory approval from the United States Food and Drug Administration (FDA) for a recombinant protein produced via a plant cell-based expression platform.
Core Technology: The ProCellEx Platform
At the center of Protalix's operational capabilities is ProCellEx, a proprietary, plant cell-based protein expression system. The platform represents an alternative to conventional mammalian (such as Chinese Hamster Ovary / CHO cells), bacterial, and yeast-based expression systems.
Key Characteristics of ProCellEx:
- Plant-Based Cell Lines: ProCellEx utilizes genetically modified tobacco (Nicotiana tabacum) and carrot cell cultures. Because plant cells do not harbor mammalian or avian viruses, the platform carries a natural biological barrier against human pathogens, eliminating viral adventitious agent risks associated with mammalian cell cultures.
- Complex Glycosylation and Post-Translational Engineering: The platform supports tailored post-translational modifications, including specific glycosylation profiles and chemical engineering (such as PEGylation), to enhance pharmacokinetic stability, prolong circulation half-life, and optimize uptake by target tissues.
- Disposable Bioreactor System: Cultivation is conducted in custom-designed, sterile, flexible polyethylene bioreactor bags. This modular setup lowers initial capital expenditure, eliminates complex clean-in-place (CIP) and steam-in-place (SIP) utility overheads, and enables horizontal scalability compliant with current Good Manufacturing Practices (cGMP).
Commercial Products
Protalix has translated its technology into multiple regulatory approvals for enzyme replacement therapies (ERT) addressing rare, hereditary lysosomal storage disorders.
1. Elelyso (taliglucerase alfa)
- Indication: Long-term enzyme replacement therapy for adult and pediatric patients with confirmed Type 1 Gaucher disease.
- Mechanism: Taliglucerase alfa is a recombinant form of the human lysosomal enzyme beta-glucocerebrosidase. It hydrolyzes glucocerebroside into glucose and ceramide, counteracting lipid accumulation in macrophages (Gaucher cells) that causes hepatosplenomegaly, cytopenias, and skeletal fragility.
- Regulatory Milestone: Approved by the U.S. FDA in May 2012, marking the first FDA approval of a human therapeutic protein derived from plant cells. It has also received approvals from ANVISA (Brazil), the TGA (Australia), and other international health authorities.
- Commercialization: Partnered with Pfizer for global commercialization, while Protalix maintains commercial rights and supply arrangements in selected territories such as Brazil.
2. Elfabrio (pegunigalsidase alfa / PRX-102)
- Indication: Long-term enzyme replacement therapy for adult patients with Fabry disease.
- Mechanism: Elfabrio is a recombinant, plant-derived human alpha-galactosidase-A enzyme that is chemically modified via a proprietary PEGylation cross-linking process. This configuration yields a stable homodimer with an extended enzymatic half-life in the bloodstream, enabling sustained clearance of globotriaosylceramide (Gb3) from the vasculature and renal endothelial tissues.
- Regulatory Status: Approved by both the U.S. FDA and the European Medicines Agency (EMA) in May 2023.
- Commercial Partnership: Co-developed and commercialized in global markets through a partnership with Chiesi Global Rare Diseases (a business unit of Chiesi Farmaceutici). Protalix conducts active cGMP drug substance manufacturing at its facility in Carmiel, Israel.
Research and Development Pipeline
Protalix leverages its proprietary manufacturing platform to engineer second-generation therapeutics for metabolic, inflammatory, and orphan indications with high unmet medical needs:
- PRX-115 (Recombinant PEGylated Uricase): An engineered plant-derived uricase enzyme under clinical evaluation for the treatment of severe, chronic refractory gout. It converts insoluble uric acid into soluble allantoin, aiming to lower serum urate levels rapidly while minimizing immunogenicity.
- PRX-119 (Long-Acting Deoxyribonuclease I / DNase I): A plant-expressed human DNase I designed to degrade extracellular DNA networks (neutrophil extracellular traps, or NETs) that accumulate in pathological concentrations during acute systemic inflammation, acute respiratory distress, and certain autoimmune conditions.
- Early-Stage and Exploratory Biologics: Ongoing discovery programs aimed at expanding the therapeutic scope of plant cell expression into additional rare metabolic enzymes and targeted biotherapeutic proteins.
